چکیده

Trypanosomes are unicellular protozoa and members of the order kinetoplastida, a highly divergent branch of the eukaryota which probably separated from the mammalian lineage ~3 x 10 years ago. Pathogenic trypanosomatids include the causative agents for Chagas’ disease or South American trypanosomiasis and Kala Azar or Leishmaniasis. They are equally divergent from the Apicomplexan protozoa that cause Malaria, Toxoplasmosis and Tularemia.

RNAi technology: A Novel approaches against fungal infections

Despite the introduction of new antifungal agents, resistances to antifungal therapy continue to increase and outcome of invasive fungal infections treatment is frequently suboptimal. A large amount of the recent effort in antifungal drug discovery has focused on a limited set of targets with functions known or expected to be important for fungal viability and virulence. A variety of techniques...

Antisense and RNA interference approaches to target validation in pain research.

In recent years numerous potential new targets for the development of novel drugs to treat chronic pain have been identified. Antisense strategies provide a straightforward approach to validate and further analyze their function. Furthermore, they offer the possibility of investigating a single member of a closely related family of proteins that cannot be easily targeted using small-molecule ph...

Down-Regulation of sidB Gene by Use of RNA Interference in Aspergillus nidulans

Background: Introduction of the RNA interference (RNAi) machinery has guided the researchers to discover the function of essential vital or virulence factor genes in the microorganisms such as fungi. In the filamentous fungus Aspergillus nidulans, the gene sidB plays an essential role in septation, conidiation and vegetative hyphal growth. In the present study, we benefited from the RNAi strate...

Genomic filtering: an approach to discovering novel antiparasitics.

Genomic filtering is a rapid approach to identifying and prioritizing molecular targets for drug discovery. For infectious disease applications, comparative genomics filters allow the selection of pathogen-specific gene products, whereas functional genomics filters, such as RNA interference (RNAi), allow the selection of gene products essential for pathogen survival. The approach is especially ...

Discovery of novel targets with high throughput RNA interference screening.

High throughput technologies have the potential to affect all aspects of drug discovery. Considerable attention is paid to high throughput screening (HTS) for small molecule lead compounds. The identification of the targets that enter those HTS campaigns had been driven by basic research until the advent of genomics level data acquisition such as sequencing and gene expression microarrays. Larg...
